何首乌中多糖的提取及清除羟自由基性能研究

摘要: 本实验探讨了何首乌多糖的提取纯化及其粗多糖对羟自由基的清除能力。考察了影响何首乌多糖的提取的工艺条件,分别对提取料液比,提取温度,提取时间进行了研究。在此基础上利用响应面分析法确定最佳工艺条件为,料液比1:12,温度为84℃,时间为105min,提取次数两次。此条件下多糖得率可达19.14%。而后,测定了经初步纯化的粗多糖清除羟基自由基功能,得到其IC50为0.27mg/ml。

Abstract: Extraction and scavenging capacity of hydroxyl radical polysaccharide from Polygonum multiflorum Thunb were studied in this paper. The factors affecting the extraction of polysaccharide were investigated. Based on the single factors test, factors such as material to solvent ratio, extracting temperature and extracting time were optimized by orthogonal test. The optimum results are: material to ratio 1:12, temperature 84 ℃ and time 105 min. On these conditions the polysaccharides yield is up to 19.14%. The experimental results demonstrated that polysaccharide can scavenge hydroxyl radical, and IC50 is 0.27 mg/ml.
