苯并[a]芘对细胞DNA修复基因XPA、XPC表达水平的影响

摘要: 目的:对食品污染物苯并[a]芘(BaP)所致细胞DNA修复基因XPA、XPC表达水平的影响进行研究。方法:用苯并[a]芘(1、5、10、50、100μmol/L)对A549染毒24h,MTT法测定细胞活性,逆转录酶及实时荧光定量聚合酶链反应的方法测定DNA修复基因XPA、XPC mRNA表达水平,用溶剂二甲基亚砜处理细胞组作为未染毒细胞组。结果:随BaP染毒浓度的增加,A549细胞活性呈下降趋势;A549细胞XPA、XPC mRNA表达水平与未染毒细胞相比,有显著降低。结论:食品污染物BaP对细胞DNA修复基因XPA、XPC mRNA表达水平有一定抑制作用。

Abstract: Objective:To investigate the effects of benzo [a] pyrene(BaP) on expression of XPA and XPC genes involved in DNA repair of A549 cells. Methods:A549 cells were exposed to different concentrations of benzo[a]pyrene(1,5,10,50 and 100μmol/L BaP) for 24 h. Cell survival was measured using MTT assay,and mRNA expression levels of XPA and XPC genes of the cells were detected using real-time quantitative RT-PCR. Results:A549 cells growth rates significantly decrease growth rate compared with the group not exposed to BaP. And the mRNA expression levels of XPA and XPC genes significanty decreased compared with the group not exposed to BaP. Conclusion:Benzo[a]pyrene could repress mRNA expressions of XPA and XPC genes in DNA repair of A549 cells.
