摘要: 本实验建立了快速分析牛血清中具有免疫活性IgG含量的方法。通过实验,确定火箭电泳法的最佳实验条件为:抗体浓度为3.0%(V/V),采用pH8.6巴比妥钠-HCl缓冲系统,用5%的戊二醛-巴比妥钠将样品1:4(V/V)醛化,非特异性蛋白用0.01mol/L pH7.4的PBST洗脱。结果表明,该方法线性结果良好,相关系数(r)为0.9947,操作简单,成本较低,适用于工业生产过程中的检测。

Abstract: The study aimed to establish a rapid method,rocket electrophoresis(RE) to detect content and activity of IgG from bovine serum. The results showed that the optimal conditions of RE are as follows:3.0%(V/V) antibody,adopting barbital sodium-HCl buffer(pH 8.6),hydroformylation with glutaraldehyde-barbital sodium(5%) at the proportion 1:4(V:V),and elution of non-specific protein with 0.01 mol/L PBST(at pH 7.4). RE is an effective way with good linearity results,correlation coefficient 0.9947,and also a simple and low-cost method,which can be used for detecting IgG during practical industrial product process.
