溴酸钾的毒理学研究

摘要: 目的:通过探讨溴酸钾对小鼠骨髓嗜多染红细胞、骨髓细胞染色体和精子的致突变性,阐明溴酸钾的毒性。方法:选用50只昆明小鼠随机分成5组,雌雄各半,设溴酸钾1/2 LD50、1/4 LD50、1/8 LD50三个剂量组,阴性对照组给予等体积蒸馏水,阳性对照组腹腔注射40mg/kg bw环磷酰胺,连续灌胃4d,观察溴酸钾对小鼠骨髓嗜多染红细胞微核率的影响;选用40只昆明小鼠随机分5组,雌雄各半,设溴酸钾1/2 LD50、1/4 LD50、1/8 LD50三个剂量组,阴性对照组给予等体积蒸馏水,阳性对照组腹腔注射40mg/kg bw环磷酰胺,连续灌胃2d,观察溴酸钾对小鼠骨髓细胞染色体的畸变情况;选用40只昆明种雄性小鼠随机分5组,设溴酸钾1/2 LD50、1/4 LD50、1/8 LD50三个剂量组,阴性对照组给予等体积蒸馏水,阳性对照组腹腔注射40mg/kg bw环磷酰胺,连续灌胃5d,观察溴酸钾对小鼠精子的影响。结果:溴酸钾的各个剂量组小鼠骨髓细胞微核率、骨髓细胞染色体畸变率及小鼠精子畸形率,与阴性对照组相比,有极显著差异(p<0.01)。结论:溴酸钾可产生明显的致突变作用,并是染色体断裂剂以及可引起精子畸形。实验结果提示溴酸钾具有潜在遗传毒性。

Abstract: Objective:Through the discussion of mutagenicity of mice born marrow polychromatic erythrocyte,born marrow cell chromosome and sperm shape abnormality induced by potassium bromate to expound its toxicity. Methods:50 Kunming mice( half of female and male ) were randomly divided into five groups:1/2 LD50,1/4 LD50 and1/8 LD50,the three dosage groups as well as negative control and positive control. The negative control group was given equal-volume distilled water,and the positive control group was conducted by abdominal cavity injection 40 mg cyclophosphamide /kg bw. The three dosage groups were conducted by gavage for four days to observe effects on frequency of mice born marrow polychromatic erythrocyte by potassium bromate. 40 Kunming mice(half of female and male) were randomly divided into five groups,and the negative control group and the positive control group were treated as the above method. Effects of potassium bromate on mice marrow cell chromosome distortion situation were observed by gastric administration to the three dosage groups for two days. To observe effect of potassium bromate on mice sperm,grouping of mice and establishment of model were the same as the experiments on mice marrow cell chromosome distortion situation basically,except gastric administration for five days. Results:Compared with negative control group,there are extremely significant differences on frequency of mice born marrow polychromatic erythrocyte,born marrow cell chromosome and sperm shape abnormality of the three dosage groups(p<0.01). Conclusions:Potassium bromate can induce obvious mutagenic action. It is a chromosome clastogen and can cause teratospermia. The results showed that potassium bromate has potential inherent toxicity.
