单环刺螠废弃内脏中粗多糖的提取工艺研究

Study on the crude polysaccharidse extracting conditions of Urechis unicinctus viscera

摘要: 利用热水浸提的方法进行单环刺螠(海肠)废弃内脏中多糖的提取。采用单因素试验和L9(34)正交试验设计,研究了不同pH、料液比、提取时间和提取温度对单环刺螠内脏多糖提取率的影响。并采用Fenton体系测定单环刺螠内脏多糖对在羟自由基(?OH)的清除作用。单环刺螠内脏多糖提取的最佳工艺条件为pH 10,料液比1:40,时间3h,温度60℃。单环刺螠内脏多糖在浓度为50μg/ml 时,对羟基自由基清除能力可达60%,且具有剂量效应。

Abstract: The crude viscera polysaccharides were extracted from Urechis unicinctus viscera by hot water extraction method . The effect of each factor such as the pH, the ratio of material to liquid, the time and the temperature on the extraction of the water-soluble polysaccharides in Urechis unicinctus viscera were investigated.using the methods of single factor test and orthogonal design test of L9(34).The scavenging action of Urechis unicinctus viscera polysaccharide on hydroxyl radical(?OH) was tested by Fenton system The results showed that the optimum extraction conditions were as follows: pH10, solid-liquid ratio 1:40, extracting time 3 h, extracting temperature 60 ℃. The scavenging capability of the crude polysaccharides to hydroxyl radical was reached to 60% at the concentration of 50 μg/ml and has dose effect.
