羊肚菌水溶性多糖PMEP-1降血脂作用研究

Hypolipidemic Activity of Water Soluble Polysaccharide PMEP-1 from Morchella esculenta (L.) Pers

摘要:

通过选用与血脂代谢及肥胖、肿瘤细胞增殖相关的PPARs 模型对羊肚菌多糖PMEP-1 的生物活性进行高通量药物筛选(high-throughput dcreening,HTS),结果发现:PMEP-1 对PPARδ模型有一定的激活作用,但作用很弱,效果不显著;对PPAR α模型没有激活作用。对羊肚菌多糖PMEP-1 降血脂作用进行动物实验,结果发现:各剂量组(低剂量100mg/kg·d、中剂量200mg/kg·d、高剂量400mg/kg·d)大鼠血清甘油三酯(TG)显著地低于高脂模型组(P<0.05),低、中剂量组大鼠血清低密度脂蛋白胆固醇(LDL-C)和动脉粥样硬化指数(AI)显著地低于高脂模型组(P<0.05),而对大鼠血清总胆固醇(TC)和高密度脂蛋白胆固醇(HDL-C)没有显著影响,说明PMEP-1 具有降血脂功能,对高脂血症的发生有一定的预防作用,同时对冠心病及动脉粥样硬化也有一定的预防作用。

Abstract:

The possible biological activities of a water soluble polysaccharide from Morchella esculenta (L.) Pers. named PMEP-1 were tested with peroxisome proliferator-activated receptor (PPAR) models related to blood lipid metabolism, and proliferation of tumor cells by high-throughput screening (HTS). PMEP-1 had slight activation effects on PPARδ models but no effects on PPAR α models. Animal experiments showed that TG in experiment groups (low dose 100 mg/kg·d, medium dose 200 mg/kg·d and high dose 400 mg/kg· d) was significantly (P< 0.05) lower than that in hyperlipidemic model group, the LDL-C content and the AI in low- and medium-dose groups were significantly (P< 0.05) lower than those in hyperlipidemic model group, while no significant differences in TC and HDL-C were seen among all the experiment groups and hyperlipidemic model group. So we concluded that PMEP-1 has hyperlipidemic acticity, and can prevent hyperlipidemia, coronary disease and atherosclerosis.
