中国林蛙蛙皮肽抗菌性质及其机理的研究

Study on Antimicrobial Activity and Mechanism of Peptide from Rana chensinensis Skin

摘要:

本实验对从中国林蛙的皮肤中提取的蛙皮肽的抗菌性质及其机理进行了研究。结果表明,该蛙皮肽抗菌谱较广,对革兰氏阴性菌、革兰氏阳性菌、酵母菌都有较好的抗菌效果,其抗菌活性具有很强的热稳定性,且在酸性条件下抗菌活性强,当pH 值大于8 时丧失抗菌活性。对经蛙皮肽处理的枯草杆菌作电镜扫描检查,发现细菌细胞壁出现破损或穿孔,而对照组细菌壁结构完整,表明该蛙皮肽的抗菌机制是通过破坏细菌细胞壁,使细胞质外流而致菌体死亡。

Abstract:

The peptide from Rana chensinensis skin has broad antibacterial spectrum, and may inhibit Gram-negative, Grampositive bacteria and yeast. The antibacterial activity also shows fairly strong thermal and acid stabilities. But when pH value is above 8, the antibacterial activity disappears. Compared with the intact cell wall of Bacillus subtilis in the control group, the broken and perforated phenomena of cell wall of Bacillus subtilis treated with the peptide was detected with scanning electron microscopy (SEM), which suggested that the antibacterial mechanism of the peptide from Rana chensinensis skin is to damage bacterial wall and result in cytoplasm outflow and bacteria death.
