双水相法提取木聚糖酶的初步研究

摘要:

采用双水相法从黑曲霉AS3.4309 的发酵液中提取木聚糖酶,通过正交试验对双水相的组成进行优化。结果表明,双水箱体系最佳组成为:选用PEG4000,其浓度为19%(W/W),磷酸氢二钾浓度为10%(W/W),氯化钠浓度为0(W/W)。此条件下,木聚糖酶的提取效果较好,分离系数(K)和上相产率(Yt)分别为6.23 和88.67%。

Abstract:

Orthogonal tests were designed to optimize the aqueous two-phase extraction process of xylanase from fermentation broth of Aspergillus niger AS3.4039 in this study. The results indicated that the optimum concentrations of PEG 4000, K2HPO4 and NaCl are 19%, 10% and 0 (W/W), respectively. Under these conditions, the separation factor (K) and xylanase yield in top phase (Yt) are 6.23 and 88.67% respectively, and the purification effect is satisfying.
