枳实提取物清除·OH 能力及抑菌活性初探

Preliminary Study on Hydroxyl Free Radical Scavenging Activity and Antimicrobial Effect of Fructus Aurantii Immaturus Extracts

摘要:

本实验研究了枳实副产物提取物的分离、纯化工艺,并对所得级分的·OH 清除作用和抑菌活性进行了初步探讨。结果表明:上样液pH 调至7.5 后再上柱,用pH6.0 浓度为70% 的乙醇溶液洗脱时大孔树脂柱分离柠檬苦素的效果最好。依次用50% 和70% 乙醇溶液洗脱Sephadex LH-20 柱可将粗提物分成5 个级分FA1~FA5,其中FA4 中含柠檬苦素9.96%。FA1~FA4 对·OH 具有不同程度的清除作用。FA1~FA5 对细菌具有一定抑制作用。

Abstract:

The isolation and purification of extracts from Fructus Aurantii Immaturus by-products after alkaloid extraction were studied, and the antioxidant activities and antimicrobial effects of different extracts were also preliminarily explored. The results showed that the optimum separation conditions for limonin by macroporous resin are as follows: sample pH 7.5, mobile solventpH 6.0, and ethanol concentration 70% (V/V). Five fractions (FA1 to FA5) can be successively obtained by eluting Sephadex LH-20 column with 50% and 70% ethanol, and the content of limonin in FA4 is 9.96%. Moreover, the fractions FA1 to FA4 show certain scavenging effects on hydroxyl radical (·OH) and all the five fractions have antimicrobial effects against bacteria.
