摘要:

采用纤维素和葡聚糖凝胶柱层析法分离纯化,获得绞股蓝多糖主要组分GPPⅢ -a,采用原子力显微镜(AFM)对GPP Ⅲ -a 的分子形貌进行观测。结果显示:GPP Ⅲ -a 分子在溶液中呈大小不一的不规则“峰”状聚集体,加热后又呈现直径大小不一的“环”状聚集体;这些“峰”状或“环”状的结构与温度等外界因素有关。

Abstract:

GPP Ⅲ -a, a main constituent of Gynostemma pentaphyllum polysaccharides (GPP), was obtained by DEAE-52 cellulose column chromatography and proved to be homogeneous by Sephadex G-150 column chromatography. The molecular morphology of GPP Ⅲ -a was observed under an atomic force microscope (AFM). The results showed that the GPP Ⅲ -a molecules were of irregular “peak”-like aggregates in solution which were changed to “ring”-like aggregates with different diameters after heat; the “peak”- and “ring”-shaped structure was associated with external factors including temperature.
