Dot Immunogold Filtration Assay for Rapid Detection of Chlortetracycline Residue in Meat

摘要: 研究斑点金免疫渗滤法快速检测禽肉中金霉素残留,将制备的金霉素多克隆抗体点在硝酸纤维素膜上,用以竞争捕获胶体金标记的金霉素-牛血清白蛋白结合物和样品中的游离金霉素,通过可见的颜色深浅判断结果。结果表明,本方法的检测限为0.4μg/ml,在金霉素浓度为0.6~1μg/ml范围内可实现定量分析。结果与酶联免疫方法的测定结果一致(相关系数r为0.970)。该法可应用于禽肉中金霉素残留的快速检测。

Abstract: A rapid detection method for chlortetracycline (CTC) was developed by dot immunogold filtration assay (DIGFA). The anti-CTC antibody was coated onto nitrocellulose membrane to capture gold-labeled CTC-BSA conjugate and CTC in samples. Positive or negative can be identified by shade of a visible red dot. Results showed that the detection limit is 0.4μg/ml. Quantitative analysis of CTC can be achieved between 0.6μg/ml and 1μg/ml. There is a good correlation of detection results between the method and ELISA with correlation coefficient of 0.970. DIGFA can be applied to detect the CTC residue in meat rapidly.
