β-胡萝卜素异构体的定性分析

摘要: 本研究发展并改进了以C30柱分离检测β-胡萝卜素异构体的高效液相色谱(HPLC)方法。在甲醇-MTBE-水(50:45:5,V/V/V)为流动相、检测波长为455nm、流速为1ml/min的色谱条件下,β-胡萝卜素熔融样品中至少有8种物质在10min内得到显著分离。借助液相色谱-大气压化学电离源质谱联用(HPLC-APCI-MS)法检测这8种物质被认为可能是β-胡萝卜素的异构体,通过比较它们的光谱特征和Q值,这些物质被依次鉴定为:15-顺-、13-顺-、9,15-双顺-、9,13-双顺-、全反式-、13,15-双顺-、9-顺-和9,13’-双顺-β-胡萝卜素。

Abstract: A developed high performance liquid chromatography (HPLC) method was introduced to determine the isomers of β-carotene. As a ternary solvent system of methanol-methyl t-butyl ether (MTBE)-water (50:45:5, V/V/V) was used as mobile phase, at lesat eight products of β-carotene melts were resolved within 10 mins with a C30 column of 1 ml/min flow rate at 455nm detection. Then these products were determined as possible isomers of β-carotene with the help of HPLC-APCI- MS. They are identified in the order, 15-cis-, 13-cis-, 9,15-di-cis-, 9,13-di-cis-, all-trans-, 13,15-di-cis-, 9-cis- and 9,13’-di-cis- β-carotene on the bases of spectral characteristics and Q ratios.
