高效液相色谱法测定小米中维生素B1

摘要: 研究反相高效液相色谱法分离测定我国主要粮食作物小米原粮中VB1的方法。小米中的VB1经前处理的酸解提取、酶解、氧化衍生、提纯后,可与小米中的淀粉、脂肪、蛋白质等物质完全分离。采用VP-ODSC18柱,以甲醇:乙酸钠(0.5mol/L,pH4.5)=20:80作流动相分离,用荧光检测器作定量测定,方法高、中、低三个水平浓度样品回收率(n=4)91.59%~94.27%;6次重复性实验相对标准偏差(RSD%)为2.8%~5.9%;方法检出限(S/N=3)为1.56ng/ml。

Abstract: A method for the determination of vitamin B1 (VB1) in millet which is one of important grain in china by high performance liquid chromatography (HPLC) was developed. After extracting by hydrochloric acid and taka diastase, the enzymatically treated sample can separated from fecula, fatty compound and protein and so on in millet completely. A VP-ODS C18 column was used for separation and elution was performed with methanol and sodium acetate (0.5 mol/L, pH4.5, 20:80). VB1 was detected by fluorescence detector quantificationally. The average recoveries were 91.59%~94.27% of high, middle and low three level (n=4)in this method. The relative standard deviations(RSD%) were 2.8%~5.9%(n=6). The detection limit (S/N=3) was 1.56 ng/ml.
