摘要: 本实验对反相高效液相色谱示差折光检测法测定蜂蜜中的果糖、葡萄糖、蔗糖和麦芽糖的方法进行了详细地探讨,结果表明:均匀称取约2.5g样品,加30ml水溶解后,再用乙腈定容至50ml,既能够有效地提取蜂蜜中的糖类物质,又保证了和流动相的相对一致性。同时对样品溶解时的分层机理作了初步推测。用该方法处理所得的样品稳定性高,测试结果重复性好。该法加标回收率均在97%~103%之间,相对标准偏差均在0.8%~1.6%之间。采用本研究方法测试了8种不同蜂蜜样品,均获得满意结果。

Abstract: This study discussed the method for determination of fructose, glucose, sucrose and maltose content in honey by reverse-phase high performance liquid chromatogram refractive index detector (HPLC-RID). Results indicated that not only the carbohydrate can be extracted from the honey effectively, but also the mobile phase relative coherence can be guaranteed by dissolving sample (2.5 g) with distilled water (30 ml) and then diluting to 50 ml with acetonitrile. Soluble mechanism was also presumed. The excellent stability and repeatability are obtained by the developed pretreatment method. The spiked recoveris is in the range of 97%~103% with relative standard deviation of 0.8%~1.6%. Eight different honey samples were detected and good results are obtained.
