响应面法优化油菜花粉谷蛋白酶解条件

Optimizing Conditions for Enzymolysis of Crude Peptide from Rape Pollen by Response Surface Methodology

摘要: 本研究在底物浓度、酶底比、pH值、酶解时间和温度五个单因素试验的基础上,利用响应面分析法对油菜花粉谷蛋白最佳酶解条件进行了研究。结果表明,Alcalase碱性蛋白酶水解花粉谷蛋白的最佳水解条件为底物浓度6%,pH值9.0,水解温度50℃,酶底比1460U/g蛋白,水解时间2h。

Abstract: Based on single-factor tests analysis (substrate concentration, enzyme concentration and substrate concen- tration ratio, pH, enzymolysis time and enzymolysis temperature), the optimum conditions of alcalase enzymatic hy- drolysis are determined by response surface methodology as follows: pH9, hydrolyzing temperature 50 ℃, enzyme concentration 1460 U/g pro substrate concertration 6%, hydrolyzing time 2 h.
