急性酒精摄入对小鼠血清抗氧化系统的影响及银杏黄酮的保护作用

摘要: 目的:研究急性酒精摄入对小鼠血清抗氧化系统的影响及银杏黄酮的保护作用。方法:昆明种成年雄性小鼠经口给予96mg/kgbw的银杏黄酮,0.5h再给予4.8g/kgbw的乙醇,同时设正常对照组和酒精对照组(乙醇4.8g/kgbw)。各组小鼠分别于末次灌胃0.5、1、2、4、6、9、15h后断头取血,测定血清谷胱甘肽(Glutathione,GSH)、谷胱甘肽过氧化物酶(Glutathioneperoxidase,GSH-Px)、超氧化物岐化酶(Superoxidedismutase,SOD)及丙二醛(Malondialdehyde,MDA)的活性或含量。结果:小鼠一次性摄入大剂量的乙醇,GSH水平及SOD、GSH-Px活性在1h后迅速下降,4h时降至最低点,随后迅速恢复至正常水平;血清MDA水平在1h后迅速上升,4h时升至最高点,15h时才缓慢恢复至正常水平。经银杏黄酮干预后,血清GSH-Px、SOD活性与GSH、MDA水平的变化曲线与酒精对照组基本相似,但GSH、GSH-Px、SOD的降幅明显低于酒精对照组,MDA的升幅也明显低于酒精对照组。结论:银杏黄酮对小鼠急性酒精性氧化损伤具有一定的保护作用。

Abstract: Objective:To study the protective effects of flavonoids of ginkgo biloba on serum anti-oxidizing system induced by acute alcohol administration on mice. Methods :After being pretreated with flavonoids of ginkgo biloba (96mg/kgbw), the mice ingested alcohol (4.8g ethanol/kgbw) via i.g. and were decapitated after 0.5h, 1h, 2h, 4h, 6h, 9h, and 15h of last time of i.g respectively. Meanwhile, the normal control group and ethanol control group (4.8g ethanol/kgbw) were employed in the study. The concentration of glutathione (GSH), malondialdehyde (MDA) and the activity of glutathione peroxidase (GSH-Px) and superoxide dismutase (SOD) in the serum were determined. Result: The experiment displayed that the content of GSH and the activities of GSH-Px and SOD decreased rapidly after 1h of treatment with alcohol and dropped to the lowest level at 4h of treatment. 6h after treatment, these indexes returned to the normal level rapidly. The level of MDA increased rapidly 1h after treatment and reached the climax at 4h. It returned to the normal concentration until 15h. On the whole, there were similar curves between flavonoids supplement group and alcohol control group on the indexes. However, to some extent, the supplement of flavonoid of ginkgo biloba could prohibit the rise of MDA level, while the decline of GSH-Px, SOD, GSH was much less than what were induced by acute alcohol intakes. Conclusion :Flavonoids of ginkgo biloba had some protective effects on the damage of serum anti-oxidizing system of mice induced by acute alcohol adminstration.
