酸浆宿萼水溶性多糖纯化及结构研究

摘要: 多糖粗提物经DE-52和SephadexG-200柱层析分离得到PAVFI、PAVFII-a、PAVFII-b、PAVFIII为均一的多糖。气相色谱、红外光谱分析结果表明PAVFI主要由阿拉伯糖组成,分子量为107kD。PAVFII-a主要是由木糖、果糖、葡萄糖组成,分子量为500kD。PAVFII-b主要是由鼠李糖、甘露糖、果糖组成,分子量为170kD。PAVFIII主要是由鼠李糖组成,且有以共价键连接的蛋白。四种多糖可能为β-(1,3)糖苷键的葡聚糖,且均含吡喃环。

Abstract: Being isolated by DE-52 and Sephadex G-200 gel filtration, four homogeneous polysaccharide which are PAVF I,PAVF II-a, PAVF II-b and PAVF III were obtained. Gas chromatography analysis and infrared spectra analysis showed that PAVF I is mainly composed of arabinose where the molecular weight of PAVF I is 107 kD. PAVF II-a is mainly composed of xylose, mannose, glucose where the molecular w-eight of PAVF II-a is 500 kD. PAVF II-b is mainly composed of rhamnose, mannose and fructose, the molecular weight of which is 170 kD. And PAVF III is mainly composed of rhamnose and some protein combined by covalent bond. Characterization of the four glycoconjugates by IR analysis showed that the typical absorption of polysaccharide indicates the presence of β-glycoside linkage.
