鹅红细胞Cu,Zn-SOD分离纯化及部分性质的研究

摘要: 经氯仿-乙醇分级分离,丙酮沉淀及DE-32纤维素柱层析的方法,从鹅红细胞中得到纯化的Cu,Zn-SOD。结果表明此酶的比活力为10656.14U/mg,提纯倍数为354.73,聚丙烯酰胺凝胶电泳蛋白带与活性带相对应。相对分子量约为32000,相对亚基分子量约为16000。最大紫外吸收波长为258nm,对KCN和H2O2敏感,最适pH为6~9,热稳定性在60℃以下。

Abstract: Copperzinc superoxide dismutase (Cu,Zn-SOD) from geese erythrocytes has been purified by chloroform-ethanol fractionation, acetone precipitation and DE-32 chromatography. The results showed that the specific activity of the enzyme was 10656.14Umg, and its purification factor was 354.73. This enzyme had two identical subunits. The molecular weight was about 32000, and the identical subunit molecular weigh about 16000. And the ultraviolet absorption peak of the enzyme was found at 258nm .The enzyme was sensitive to KCN and H2O2, the enzyme optimum pH was from 6 to 9, stable below 60℃.
