氯羟吡啶人工抗原合成和鉴定

摘要:

目的:将氯羟吡啶的衍生物分别与牛血清白蛋白(BSA)和鸡卵清白蛋白(OVA)制备人工抗原。方法:采用威廉森合成法合成了氯羟吡啶的衍生物(CLOP-2C),再分别经活泼酯法和氯甲酸异丁酯法合成氯羟吡啶的人工抗原CLOP-2C-BSA,CLOP-2C-OVA)。结果:通过TLC、质谱、红外光谱等方法鉴定表明成功制备了氯羟吡啶衍生物。紫外扫描图谱表明人工抗原偶联成功,且半抗原与载体的分子结合比率分别为8.26 和4.7。结论:成功的制备了氯羟吡啶人工抗原。

Abstract:

Objective: The artificial antigens were synthesized by derivative of clopidol and BSA、OVA.Methods: The derivative of clopidol, called as CLOP-2C, was synthesized by Williamson method in this study. Then two artificial antigens of clopidol(CLOP-2C-BSA, CLOP-2C-OVA) were prepared by active ester method (AE) and mixed anhydride method (MA) respectively. Results: CLOP-2C is identified by TLC, MS, IR spectrum . The characterization of hapten-protein conjugates were examined by ultraviolet spectrophotometry. The numbers of hapten molecules per molecule of protein estimated by ultraviolet spectrophotometry according to the absorbance of compounds at 280 nm for two conjugates were 8.26 and 4.7 respectively Conclusion: The artificial antigens were synthesized successful.
