中国林蛙皮抗菌肽提取条件的优化研究

Optimization of Extraction Conditions for Antimicrobial Peptides from Skin of Rana chensinensis

摘要: 本研究以林蛙干皮为原料,采用甲醇、乙酸和磷酸盐三种浸提液提取有抑菌活性的多肽,并分别以提取温度、时间、液料比、pH值四因素对每种浸提液的提取方法进行三水平正交试验,以进一步精纯后产物的抑菌活力为指标,探索林蛙抗菌肽的最优分离纯化工艺。结果表明,三种浸提液的最优条件分别是:甲醇法为浸提时间24h、温度0℃、液料比为22.5(W/V)、pH5.4;乙酸法为24h、0℃、液料比22.5、pH3.0;磷酸盐法为24h、20℃、液料比22.5、pH6.4。其中甲醇法浸提产物的抑菌活性最佳,粗提品经G-25层析后,力活提高了5倍,对大肠杆菌和金黄色葡萄球菌均有抑制作用。

Abstract: In this study,methanol,acetic acid and phosphate solution were used for extracting antimicrobial peptides from dry skin of Rana chensinensis respectively. For the three extraction processes,the important process parameters such as temperature,time,extraction solution to material ratio and pH were optimized respectively by using orthogonal test L9(34) with antibacterial activity of the purified product as test index. The results showed that the optimal conditions of the methanol extraction method are 0 ℃,24 h,extraction solution to material ratio 22.5:1,and pH 5.4; For the acetic acid extraction method,they are 0 ℃,24 h,extraction solution to material ratio 22.5:1,and pH 3.0; For the phosphate solution extraction method,they ar 20 ℃,24 h,extraction solution to material ratio 22.5:1,and pH6.4. Among the three methods,the antibacterial activity of the extract obtained by the methanol extraction method is the highest. The specific antibacterial activity is increased by about 5 times through Sephadex G-25 chromatography. The purified product has some antibacterial effects against both Staphylococcus aureus and Escherichia coli.
