板党多糖酶法提取研究

摘要: 本实验将现代酶解技术应用于板桥党参多糖的提取,由单因素及正交试验所确定的木瓜蛋白酶提取板党多糖的最优工艺参数为酶用量60U/g干粉、酶解时间90min、pH5.0、温度40℃,此条件下的多糖得率为23.73%;纤维素酶提取板党多糖的最优工艺参数为酶用量120U/g干粉、酶解时间150min、pH4.0、温度40℃,此条件下的多糖得率为28.22%。

Abstract: In this experiment,the modern enzymolysis technology was applied for extraction of polysaccharide from Banqiao Codonopsis pilosulc.Based on single factor tests,the extraction conditions of polysaccharide were optimized by orthogonal test.The results of orthogonal test showed that the optimal extraction conditions of polysaccharide by papain are as follows:enzyme amount 60 U/g dry powder,enzymolysis time 90 min,pH 5.0 and temperature 40 ℃.Under these conditions,the percent of extraction of polysaccharide is 23.73%.For cellulase,the optimal extraction conditions are as follows:enzyme amount 120 U/g dry powder,enzymolysis time 150 min,pH 4.0 and temperature 40 ℃.Under these conditions,the percent of extraction of polysaccharide is 28.22%.
