轮梗霉高产花生四烯酸的诱变育种研究

Study on Arachidonic Acid Mutation Strains Breeding of Diasporangium sp.

摘要: 利用紫外线作为诱变剂,分别通过15℃的低温和0.09mg/ml乙酰水杨酸的筛选作用,筛选到两株花生四烯酸产量有较大提高的菌株,分别为LT1.12菌株(花生四烯酸产量提高46.41%)和A1.13菌株(花生四烯酸产量提高53.15%)。通过对脂肪酸组分的分析,确定乙酰水杨酸为更加具有针对性的筛选条件。并对用乙酰水杨酸筛选到的A1.13菌株进行了传代稳定性实验,证实A1.13菌株花生四烯酸产量提高这一性状是可以稳定遗传的。

Abstract: Using Diasporangium sp. as original strain, with low temperature 15℃ and 0.09 mg/ml aspirin as inhibitors, after UV radiation, some mutation strains were selected. Fermentation experiments assayed various strains in the biomass, arachidonicacid yield and fatty acids. The changes of fatty acids showed that the aspirin is the most effective inhibitor, and A1.13 strain is selected with its arachidonic acid yield enhanced 53.15%.
