改良热酚水法制备大肠杆菌O157:H7脂多糖抗原的研究

摘要: 采用热酚水法提取肠出血性大肠杆菌(EHEC)O157:H7的脂多糖(LPS),经浓缩酶解,离心分离得纯化LPS,酸解制得O-特异性侧链(O-PS),能有效去除核酸及杂蛋白,抗原具有鲎试剂凝集活性及对产蛋鸡有较强的免疫原性,建立了一种改良热酚水法提取制备O157:H7特异性脂多糖抗原的方法。

Abstract: The LPS antigen was prepared from EHEC O157:H7 strain 933 with hot phenol water method and purified by enzyme hydrolyzing with DNase and RNase, and then hydrolyzed by 1.5% acetic acid for getting OPS. The O157:H7 LPS antigen has high purity specificity, has activity of agglutinating limulus amebocyte lysate and high immunoreactivity for chicken. They could be used to produce specific anti-LPS antibody.
