醋酸甲羟孕酮完全抗原制备及ELISA检测

摘要: 本实验采用碳二亚胺法合成完全抗原MPA-BSA,并采用紫外分光光度法及SDS-PAGE凝胶电泳鉴定对偶合物进行鉴定,使用合成抗原对兔子进行免疫,得到抗血清,在此基础上建立醋酸甲羟孕酮酶联免疫检测方法,检测限为1.8μg/ml,线性范围为2.5~50μg/ml(r2=0.9936)。

Abstract: Medroxy progesterone acetate(MPA), as a hapten, cannot be led to immune reaction and has to be conjugated with a carrier, such as bovine serum albumin(BSA). The carbodiimides reaction was used to prepare the immunogen in this paper. The conjugates of MPA-BSA were identified by the analysis of UV-spetrum, SDS-PAGE and animal immunization test. An ELISA method was established with MPA antibody, which has the detection limit of 1.8μg/ml and the linear detection of 2.5~ 50μg/ml(r2=0.9936).
