摘要: 为探讨不同饲料构成对大鼠褐色脂肪组织解偶联蛋白-1(UCP1)、白色脂肪组织解偶联蛋白-2(UCP2)和骨骼肌解偶联蛋白-3(UCP3)基因表达的影响,利用不同的饲料(包括基础饲料、高蛋白、高脂肪、高能1与高能2饲料)喂养雄性Wistar大鼠2个月,观察不同饲料构成对大鼠体重、脂体比的影响,并应用RT-PCR方法测定UCP1、UCP2和UCP3mRNA表达水平。结果表明,高能饲料1组与高能饲料2组的体重、脂体比均显著高于基础组、高蛋白组与高脂肪组(p<0.05),而后三者的体重、脂体比无显著性差异(p>0.05)。高能饲料1组与高能饲料2组UCP1、UCP2基因表达增强,UCP3基因表达降低,能量相同的基础组、高蛋白组与高脂肪组没有明显差异。说明能量能诱导大鼠褐色脂肪组织UCP1和白色脂肪组织UCP2基因表达,但降低骨骼肌UCP3基因的表达。

Abstract: In order to explore the effects of different diet compositions on the expression of gene of uncoupling proteins 1 (UCP1) in rat brown adipose tissue(BAT), uncoupling proteins 2 (UCP2) in white adipose tissue(WAT) and uncoupling proteins 3 (UCP3) in skeletal muscle, male Wistar rats were fed with different diets (including basic diet, high protein diet, high fat diet, and high calorie 1st and 2nd diet) for 2 months. The effects of the different diets on weight and the ratio of fat to body weight were observed. Reverse transcription-polymerase chain reaction (RT-PCR) technique was used to measure UCP1, UCP2 and UCP3 mRNA. The results showed that the body weight and the ratio of fat to body weight in the high calorie 1st and 2nd diet group are obviously higher than that in the groups of basic diet, high protein diet, and high fat diet (p<0.05), but there is no significant difference between the last three groups (p>0.05). The expression of UCP1and UCP2 genes increases and the expression of UCP3 gene decreases in the high calorie diet 1st and 2nd group, and no significant difference between basic diet, high protein diet and high fat diet group with equaled energy is observed. Energy can induce the expression of gene of UCP1 in rat BAT and UCP2 in WAT, but decrease the expression of gene of UCP3 in skeletal muscle.
