东北山核桃仁油对D-半乳糖衰老小鼠抗氧化能力的影响

Effect of Oil from Juglans mandshurica Grown in Northeast China on Antioxidant Abilities in D-Galactose-Induced Aging Mice

摘要: 研究东北山核桃仁油(Juglans mandshurica Oil,JMO)对D-半乳糖衰老小鼠抗氧化能力的影响。采用连续颈背部注射D-半乳糖建小鼠亚急性衰老模型的方法,造模6周。建模同时,以VC为阳性物质对照组,JMO低、中、高剂量组分别灌胃5、10、20mL/(kg·d),同时设模型对照组,灌胃生理盐水。6周后测定小鼠血清、肝脏、脑组织中总抗氧化能力(T-AOC)、超氧化物歧化酶(SOD)、丙二醛 (MDA)、过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GSH-Px)和单胺氧化酶(MAO)的含量。结果表明:模型对照组血清、肝脏和脑组织匀浆中T-AOC、SOD、CAT、GSH-Px含量均显著降低,MAO和MDA含量显著升高,与正常对照组比较差异显著;10、20 mL/(kg·d) 中、高剂量JMO可显著提高衰老小鼠机体总抗氧化能力T-AOC水平,显著提高抗氧化酶SOD、CAT和GSH-Px的活性,明显降低MAO活性和脂质过氧化物MDA含量,与模型对照组比较有显著性差异。提示,JMO能抑制机体内脂质过氧化物的生成,有效清除机体内产生的过量自由基,显著提高亚急性衰老小鼠的抗氧化能力,具有量效关系。

Abstract: To investigate the influence of Juglans mandshurica Oil (JMO) on antioxidant abilities in aged mice, a subacute aging mice model was built by neck back subcutaneous injection of D-galactose for 6 consecutive weeks. Using VC as positive control, JMO was given to mice by gavage at three different doses of 5, 10 mL/(kg·d) and 20 mL/(kg·d). The control and model control groups were given normal saline by gavage during that time. Total antioxidant capacity (T-AOC), catalase (CAT), malondialdehyde (MDA), superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and monoamine oxidase (MAO) in the serum, liver and brain of mice were determined at the end of the last administration. The results showed that the content of T-AOC, SOD, CAT and GSH-Px in the aged model group significantly declined (P <0.05), but the content of MAO and MDA significantly increased as compared to the normal control group (P <0.05). For the aged mice treated with 10 mL(kg·d)and 20 mL/(kg·d) of JMO, the activities of SOD, CAT, GSH-Px and the content of T-AOC significantly increased (P <0.05) and the activity of MAO and the content of MDA significantly declined (P <0.05) as compared to aged control mice. These data indicated that JMO could inhibit the generation of lipid peroxides, and effectively remove free radicals, and significantly increase antioxidant ability in the body in a dose-dependent manner.
