黑玉米籽粒中花色苷提取条件的优化

Optimization of the Extraction Strategy for Anthocyanin Content in Kernel of Dark Maize

摘要: 本研究以西南大学新创的全黑玉米自交系SDM(Super Dark Maize)为材料,以提高花色苷得率和纯度为目的,采用正交试验研究了乙醇浓度、固液比、提取液pH值、提取时间、提取温度等五个提取条件对籽粒中花色苷得率和纯度的影响。结果表明:在90%的乙醇、固液比1:15、pH值1、在80℃保温15h的条件下,花色苷得率最高,但纯度最低;在保证花色苷纯度较高的前提下,考虑最高得率,以处理组合A3B3C2D2E3效果最佳,即得最优提取条件为:90%的乙醇、固液比1:15、pH=1.5、在70℃保温15h。

Abstract: Orthogonal experiment was carried out to reveal the influence of ethanol concentration, solid-liquid ratio, pH, extraction time, and extraction temperature related to the extraction strategy for the yield ratio and purity of extracted anthocyanin from the kernels of super dark maize (SDM). Results from the systematic ANOVA and multiple comparison showed that treatment with ethanol concentration of 90%, solid-liquid ratio of 1:15, pH value of 1, extraction temperature of 80℃, and extraction time of 15h possessed statistically higher yield ratio of anthocyanin but significant lower purity than the other treatments. Considering the relatively higher purity and yield ratio of extracted anthocyanin, an optimized extraction strategy was determined as treatment(A3B3C2D2E3) with ethanol concentration of 90%, solid-liquid ratio of 1:15, pH value of 2, extraction temperature of 70℃, and extraction time of 15h.
