响应面分析法优化β-环糊精包合分离花椒籽油中α-亚麻酸的工艺

Optimiazaion of the Separation Technique for α-linolenic acid by β-cyclodetrin inclusion from Pricklyash Seed Oil with Response Surface Methodology

摘要: 摘 要:在单因素试验的基础上,以水/β-环糊精(β-CD)、包合温度、包合时间为自变量,α-亚麻酸的提纯率为响应值,采用Box-Behnken试验设计,利用响应面分析法对花椒籽油中α-亚麻酸的分离工艺进行优化。结果表明:试验确定的最佳分离工艺条件为:水/β-环糊精(β-CD)9.9:1,包合温度64℃,包合时间1.32h,β-环糊精包合花椒籽油的包合率最高达22.423 %。通过气相色谱-质谱联用仪(GC-MS)和红外光谱仪对包合前后脂肪酸成份进行了分析,共检测出3种脂肪酸,使α-亚麻酸的相对含量由4.12%提高到21.35%。

Abstract: Abstract: The optimized separation conditions for α-linolenic acid from Pricklyash Seed Oil were determined by using the single factor experipents and response surface analysis. The ratio of water to β-CD , inclusion temperature and inclusion time were taken as independent variable and yield of α-linolenic acid was taken response value based on single factor tests. The results showed that the optimum seperation conditions were as follows that the ratio of water to β-CD was 9.9:1, addution temperature 64℃, addution time 1.32h. Inclusion rate reached 22.423%. The constitutes of unsaturated fatty acids were analyzed by GC-MS and FT-NIR after included, and there are three kinds of unsaturated fatty acids. The content of α-linolenic acid was improved from 4.12% to 21.35%.
