镉胁迫下乳酸乳球菌的形态变化

Morphology Change of Lactococcus lactis subsp.Lactis under Cadmium Stress

摘要: 摘 要:通过扫描电镜和透射电镜对不同浓度镉胁迫下培养的泡菜乳酸乳球菌的表面和亚细胞结构的变化进行研究。研究分别对培养基中含有0mg/L、10mg/L、50mg/L、100mg/L、200mg/L Cd2+的菌体细胞进行观察,结果表明泡菜乳酸乳球菌呈球形,表面光滑, 长径960nm,短径775nm(未添加Cd2+)。添加50mg/L Cd2+处理菌体细胞表面产生大量白色颗粒状物质; 添加Cd2+ 100 mg/L时菌体细胞存活数量大幅度下降,并且发现白色颗粒状物质聚集成团出现,甚至形成大块的生物岩石状。透射电镜结果显示:没有添加Cd2+时泡菜乳酸乳球菌结构完整,细胞内容物分布均匀,在10, 50mg/L Cd2+ 胁迫下菌体生长较为正常,当菌体暴露于100, 200mg/L Cd2+ 时菌体细胞出现异常现象如细胞破裂,内容物从薄膜穿孔中释放,质壁分离等。两类电镜结果均说明:在低浓度Cd2+ (≤50 mg/L)胁迫下,对泡菜乳酸乳球菌的生长几乎不产生影响,添加Cd2+质量浓度上升到100,200mg/L时泡菜乳酸乳球菌正常生长受到抑制。

Abstract: Abstract:By scanning electron microscopy (SEM)the cell face and subcellular structure in different contration of Cd were investigated.In the medium containing 0mg/L,10 mg/L, 50 mg/L, 100 mg/L, 200 mg/L Cd2 + Lactococcus lactis cells were observed .The SEM result showed that the PC strain(with no Cd2 +) was spherical and had smooth surface;the long diameter was 960 nm and the short diameter was 775 nm. When the added Cd=50 mg/L, there were many white bulkloads on the surface. The amount of Survival Stains began to fall under 100 mg/L Cd2 + and the white bulkloads aggregated to conglobation and became biolith with 200 mg/L Cd stress. The TEM results showed that PC strain structure was intact and cellular content were well distributed with no Cd2 +; the strain keep well in 10, 50 mg/L ,when exposed to 100 ,200 mg/L Cd2 + it began to change anormaly such as cell lysis and the release of cellular contens from holes in the cell membrance ,in addition , there were piasmolysis and disappear in cell wall and membrance. The total results indicated that the low concentration of Cd2 + (≤50 mg/L) have nearly no effection to PC strain and began to inhibit the normal growth of it when Cd2 + up to 100, 200 mg/L.
