解淀粉芽孢杆菌H15产抗菌肽的发酵和提取条件

Fermentation and Extraction Technology for Antifungal peptides Production by Bacillus amyloliquefaciens H15

摘要: 以前期筛选的分泌高活性抗菌肽的解淀粉芽孢杆菌(Bacillus amyloliquefaciens H15)为出发菌株,尖孢镰刀菌为指示菌,无菌滤液抑菌圈直径为考察指标,对解淀粉芽孢杆菌H15所产抗菌肽的发酵和提取条件进行优化。获得优化后的最佳培养基为L-1培养基,最佳培养条件为:接种量3%、起始pH 6.68、培养温度30.79℃、培养时间48.61h,转速160 rpm。在此优化条件下,发酵液抑菌直径可高达25.21 mm/100ul,比优化前(16.40mm/100ul)提高了52.5%。以4株玉米中携带的优势霉菌(草酸青霉、串珠镰刀菌、尖孢镰刀菌、黑曲霉)为指示菌,比较分析不同浓度硫酸铵沉淀和盐酸沉淀不同有机溶剂抽提对解淀粉芽孢杆菌H15抗菌肽提取的影响,结果均表明,盐酸沉淀丙酮抽提法活性最高。

Abstract: Fermentation and extraction conditions for antifungal peptides production from Bacillus amyloliquefaciens H15 were optimized. Inhibitory zone was used as evaluation index and quantified against Fusarium oxysporum ACCC 37438. Results showed the optimal fermentation conditions were L-1 medium, inoculum amount of 3 percent inoculum, initial pH of 6.68, rotate speed of 160rpm, fermentation time of 48.61 h and fermentation temperature of 30.79 ℃. Under these optimal conditions, the bacteriocin inhibitory activity was increased 52.50 percent and reached up to 25.21 mm/100ul. Selected four fungi induced food corruption as indicator fungi, we compared with different extraction methods of the antimicrobial substances. The results showed that the extraction method of hydrochloric acid precipitation and organic solvent extraction were better than the extraction method of ammonium sulfate precipitation method. The acetone extraction effect was significantly better than other four organic solvent .
