海湾扇贝多肽对人肝癌 HepG2细胞增殖和凋亡的影响

Effect of the Polypeptide from Bay Scallop on the Proliferation and Apoptosis of HepG2 Cells

摘要: 采用CCK-8法检测海湾扇贝多肽对体外培养的人肝癌HepG2细胞增殖的抑制作用,利用倒置显微镜及流式细胞仪观察、分析其对HepG2细胞的细胞形态和细胞凋亡的影响。结果表明,不同浓度海湾扇贝多肽(4 mg/mL、6 mg/mL、8 mg/mL、10 mg/mL和12 mg/mL)能够抑制人肝癌HepG2细胞的生长,且抑制率随海湾扇贝多肽浓度的升高而增加。经12 mg/mL海湾扇贝多肽处理后,肿瘤细胞的细胞形态发生了明显改变,表现为细胞皱缩、透明度降低、细胞核固缩等,并出现了凋亡小体。海湾扇贝多肽可诱导HepG2细胞凋亡。

Abstract: The antitumor efficiency and mechanism of the PBS were studied by using CCK-8 assays, inverted microscope and fluorescence-activated cell sorter (FACS).The results showed that the different concentrations PBS (4 mg/mL,6 mg/mL,8mg/mL,10mg/mL and 12mg/mL) could suppress the growth of human hepatocellular carcinoma cell HepG2, and the growth inhibition rates increased with the concentration of the PBS. After treated with PBS of 12 mg/mL, the morphology of two tumor cells were changed, characterized by cell shrinkage, transparency decrease and nuclear chromatin pyknosis, etc. and the apoptotic bodies were found in 12 mg/mL experimental group. HepG2 cell apoptosis was induced.
