水杨酸、茉莉酸甲酯及超声波对北沙参愈伤组织生长及生产香豆素的影响

Effects of salicylic acid, methyl jasmonate and ultrasound on the production of cumarin and growth of Radix Glehniae callus

摘要: 研究了水杨酸(Salicylic Acid,SA)、茉莉酸甲酯(Methyl Jasmonate,MeJA)和超声波对北沙参愈伤组织生长的影响,并利用高效液相色谱法(HPLC)测定了经诱导处理后愈伤组织中香豆素(欧前胡素、异欧前胡素及补骨脂素为主要指标)含量的变化。结果表明:SA和MeJA均对北沙参愈伤组织的生长具有抑制作用,而一定时间的低强度超声处理则能够促进北沙参愈伤组织生物量的增长;同时,研究结果表明此三种诱导因子均能够不同程度的促进香豆素合成,即当SA浓度达到8 mg/L时,欧前胡素的含量达到最大值,为对照的3.8倍;SA浓度为16mg/L时,补骨脂素达最大值,为对照的1.7倍;当SA浓度在28-30mg/L之间时,异欧前胡素的含量达到最大值,此范围内总香豆素含量也达最大值,约为对照的2.33倍;当MeJA浓度达400μmol/L时,补骨脂素和异欧前胡素含量达到最大值,分别为对照的6.09倍和5.53倍,总香豆素含量也达最大值14.86μg/g,为对照的5.11倍;当超声处理30 min时,愈伤组织中欧前胡素和异欧前胡素的含量达最大值5.51 μg/g和3.69μg/g,分别为对照的5.15倍和4.2倍;总香豆素含量也达最大值11.9μg/g,为对照的3.55倍;而补骨脂素则在超声处理10min后达到最大值3.6 μg/g,为对照的1.5倍。综上比较三种诱导子对北沙参愈伤组织诱导处理后香豆素含量的变化发现,与SA和超声波相比,MeJA更能够有效的促进北沙参中次生代谢产物——香豆素的合成。

Abstract: We studied the influence of Methyl Jasmonate、Salicylic Acid and Ultrasonic on the production of cumarin ( imperatorin, isoimperatorin and psoraleae) and growth of callus of Radix Glehniae. The results showed that SA and MeJA could cause the callus brown, while a certain time and strength of ultrasound had a positive effect on callus vitality. At the same time, the results showed that the production of Cumarin could be promoted by the three kinds of factors. When the concentration of salicylic acid reached 8mg / L, the content of imperatorin reached the Maximum, which was 3.8 times higher than contrast; but the psoralen had a little effect by SA, when the concentration of salicylic acid reached 16mg / L, the production reached maximum, which was 1.7 times higher than contrast; When the concentration of salicylic acid reached 28-30mg / L, The maximum value of the content of the isoimperatorin and total cumarin which was 2.33 times higher than contrast; when the concentration of MeJA reached 400 μmol / L, the production of psoralen and isoimperatorin reached maximum, which respectively was 6.09 and 5.53 times higher than contrast, and the cumarin of total which was 5.11 times higher than contrast; when the time of ultrasound was 30min, the production of imperatorin、isoimperatorin and total cumarin reached maximum,which was respectively 5.15 、4.2and 3.55 times higher than contrast; when ultrasonic treatment 10min, the production of psoralen reached maximum, which was 1.5 times higher than contrast. In summary, comparison of three kinds of elicitors of Radix Glehniae callus,which was induced in the content of coumarin was detected,compared with ultrasonic and SA, MeJA could effectively promote the production of coumarin of Radix Glehniae .
