植物乳杆菌Sc52联合牛蒡低聚果糖对糖尿病模型小鼠的治疗作用

Therapeutic Effects of Burdock Fructooligosaccharide Combined With Lactobacillus Plantarum Sc52 on Type II Diabetes Mice Model

摘要: 目的:研究植物乳杆菌(Lactobacillus plantarum)Sc52和牛蒡低聚果糖(burdock fructooligosaccharide,BFO)联合使用对高脂饲料结合链脲佐菌素(streptozocin,STZ)诱导的2型糖尿病小鼠的治疗作用。方法:高脂饲料结合STZ建立2型糖尿病小鼠模型,分别灌胃植物乳杆菌Sc52、BFO及Sc52+BFO,连续4 周。检测小鼠空腹血糖含量、空腹血糖耐受量,血清中脂质、胰岛素和炎症因子水平等相关生化指标;分析小鼠粪便中短链脂肪酸含量及主要菌群数量。结果:Sc52+BFO组合能够抑制小鼠血清中总胆固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)、低密度脂蛋白胆固醇(low density lipoprotein cholesterol,LDL-C)、内毒素(endotoxin,ET)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-6(interleukin-6,IL-6)水平的升高,明显地提升血清中高密度脂蛋白胆固醇(high density lipoprotein cholesterol,HDL-C)、胰岛素(insulin,INS)水平;Sc52+BFO组合还能提高小鼠肠道中乙酸、丙酸、丁酸含量,降低乳酸含量;促进小鼠肠道中乳杆菌、双歧杆菌的生长,抑制肠球菌、肠杆菌、产气荚膜梭菌等有害微生物的生长,且Sc52+BFO组合治疗糖尿病的效果优于单纯使用BFO和Sc52。结论:植物乳杆菌Sc52和牛蒡低聚果糖联合使用能够调节小鼠肠道菌群平衡、降低血糖血脂含量、抑制炎症因子表达,从而提高2型糖尿病小鼠胰岛素水平。

Abstract: Objective: To explore the therapeutic effects of Burdock fructooligosaccharide (BFO) combined with Lactobacillus plantarum Sc52 on type 2 diabetes induced by high-fat diet and streptozocin (STZ). Methods: C57BL/6J mice were intraperitoneally injected with fed with high-fat diet and STZ to establish a mouse model of Type 2 diabetes, then Sc52, BFO, and Sc52 combined with BFO were respectively given to mice by gavage administration for 4 continuous weeks. The fasting blood glucose and tolerance dose were measured by Glucose Test Strips; the serum lipids, INS, inflammatory factor and other biochemical indicators were also tested by kits; the contents of short-chain fatty acids were tested by GC and the main intestinal bacteria in feces were counted by coated plates. Results: L. plantarum Sc52 and BFO alone or combination could inhibit the elevation of TC, TG, LDL-C, ET, TNF-α and IL-6 in serum, significantly increase HDL-C and INS levels in serum. The contents of acetic acid, propionic acid and butyric acid in intestine were raised but the content of lactic acid was decreased. L. plantarum Sc52 combined with BFO could promote the growth of Lactobacillus and Bifidobacterium, inhibit the growth of Enterobacilli, Enterococcus, Clostridium perfringens and other harmful bacteria in the intestine of mice and the therapeutic effects of L. plantarum Sc52 combined with BFO was better than that treated by BFO or L. plantarum Sc52 alone. Conclusion: L. plantarum Sc52 combined with BFO may improve insulin in a mouse model of Type 2 diabetes by regulating the balance of intestinal flora in mice, decreasing the content of blood glucose and serum lipids and inhibiting inflammatory cytokines.
