不同干燥方法对乳源ACE抑制肽活性的影响

Effect of different drying process on ACE activity inhibition peptides from bovine casein hydrolysate

摘要: 用不同的干燥方法干燥富含ACE抑制肽的酪蛋白水解物,研究对ACE活性抑制的影响。在研究过程中,采用喷雾干燥法、流化床干燥法和真空冷冻干燥法干燥富含ACE抑制肽的酪蛋白水解产物,以干燥产物的水分含量低于5%为指标,确定干燥条件,以干燥产物的ACE活性半数抑制浓度(IC50)值,优化干燥条件。并通过动物实验,评价不同干燥方法所得产物的降血压效果。结果显示,采用喷雾干燥法干燥水解产物条件为: 进风温度190℃、出风温度75℃、进料量44 mL/min,其干燥产物的水分含量为3.851±0.037%,ACE活性抑制的IC50值为0.442mg/mL;流化床干燥水解物的条件为:进风温度65℃、进料量180mL、悬浮介质添加量30g,其干燥产物的水分含量为4.491±0.128%,ACE活性抑制的IC50值为0.294mg/mL;真空冷冻干燥产物的水分含量为1.68%,ACE活性抑制的IC50值为0.275mg/mL。动物实验结果显示,用相同剂量灌胃三种干燥方法所得干燥产物,采用真空冷冻干燥方法,有利于产物活性的保留,其次时流化床干燥,而喷雾干燥产物的活性损失最大。因此,在富含ACE抑制肽水解产物干燥时,采用冷冻干燥方法较好。

Abstract: The objective of this paper was to research effect of different drying process on angiotensin converting enzyme (ACE) activity inhibition peptide from casein hydrolysate. In the research, the methods which spray drying process and fluid bed drying and vacuum freeze drying were used to dry casein hydrolysate. Moisture content is lower than 5% as a standard to determine drying conditions, and the conditions were optimized base on half inhibitory concentration (IC50) of ACE activity. The antihypertensive effects of drying productions were evaluated by animal experiment. The results showed that, the condition of spray drying method were that inlet air temperature is 190℃, outlet air temperature is 75℃ and input quantity is 44 mL/min, the moisture content of drying production was 3.851±0.037%, and the IC50 of ACE activity was 0.442mg/mL; the drying condition of fluid bed drying method were that inlet air temperature is 65℃,inlet amount is 180 mL and additive amount of suspending medium is 30g, the moisture content of drying production was 4.491±0.128% and IC50 of ACE activity was 0.294 mg/mL; the moisture content of dried production by vacuum freeze drying was 1.68%, and the IC50 of ACE activity was 0.275 mg/mL. The results of animal experiment showed that, as spontaneously hypertensive rat (SHR) was oral administrated with drying productions by three drying methods processed in the same dose, the anti-hypertension activity of drying production by vacuum freeze drying was best keep, it was better by fluid bed drying, and it was worst by spray drying. Therefore, it was one better method using vacuum freeze drying method to drying process casein hydrolysate which rich in ACE inhibition peptides.
