不同茶类发花茯茶中“金花”菌的分离、鉴定及产黄曲霉毒素研究

Study on identification and the aflatoxins-producing capability of ‘golden flora’ fungal isolated from the Fu tea produced by the different kinds of tea

摘要: 本研究以乌龙茶(大红袍)、红茶、绿茶、黑茶(天尖原料/金湘益茯砖茶原料)五种茶样为原料,运用“散茶发花”技术制得不同茶类散装茯茶,基于各成品茶中“金花”菌间存在一定的形态差异,试验运用微生物手段对各茯茶制品中“金花”菌进行了差异分析,同时从形态与DNA序列分析角度对其进行了系统的鉴定,并运用TSQ Quantum三重四级杆串联质谱对其是否具备产黄曲霉毒素的能力进行了检测分析。结果表明:不同茶类发花茯茶成品中的“金花”菌实属同一种真菌,无异于原接种供试“金花”菌,该菌被鉴定为真菌门(Eumycota; Mycobionta)子囊菌纲(Ascomycetes)真子囊菌亚纲(Euascomycetes)曲霉目(Eurotiales)曲霉科(Eurotiaceae)散子囊菌属(Eurotium Link ex Fires)冠突散囊菌(Eurotium cristatum),无性型为针刺曲霉(Aspergillus spiculosus Blaser);此外,质谱数据显示未检测出AFB1、AFB2、AFG1和AFG2,表明该分离“金花”菌不具备产黄曲霉毒素的能力。

Abstract: It was vaccinated in different tea samples such as Oolong tea (Dahongpao), Black tea, Green tea, Dark tea (the raw material of Tianjian tea and the raw materials of Jinxiangyi brick tea) by ‘fungal fermentation with loose tea’ to obtain five kinds of Fu tea whose ‘Golden Flora’ showed differences in morphological. This study analyzed the difference of it through microbial method and made systematic evaluations from morphology and DNA sequence analysis. Meanwhile, the toxigenicity has been studied by TSQ Quantum. The results revealed that the ‘Golden Flora’ is the same fungus in five kinds of Fu tea, also the same with the one on vaccination for test, which is classified as Eumycota, Ascomycetes; Euascomycetes; Eurotiales; Eurotiaceae; Eurotium Link ex Fires; Eurotium cristatum; Aspergillus Spiculosus Blaser. And the mass spectrometry data showed that AFB1, AFB2, AFG1 and AFG2 was not detected, which indicate the ‘Golden Flora’ is not able to produce aflatoxins.
