T-2毒素对凡纳滨对虾DNA粘度损伤效应的研究

Research on T-2 toxin on damage effect of Litopenaeus vannamei DNA viscosity

摘要: 为探究T-2毒素对对虾DNA粘度的损伤效应,通过小牛胸腺DNA与T-2毒素作用,采用响应面分析其最佳作用条件,再通过体外T-2毒素与对虾DNA作用验证,最后通过喂食对虾T-2毒素进行蓄积染毒后,观察对DNA粘度的影响。结果:响应面分析优化出对DNA粘度影响最明显的反应条件为T-2毒素浓度为2.7ng/mL、DNA浓度为50μg/mL、反应时间为43min;小牛胸腺DNA和对虾DNA与T-2毒素作用会引起DNA粘度一致的变化,而剂量组T-2毒素染毒对虾体内的DNA粘度变化于体外实验的结果不完全一致。

Abstract: To explore the damaging effects on shrimp DNA viscosity of T-2 toxin, through the reaction of calf thymus DNA and T-2 toxin analyze the optimol conditions with response surface analysis . The reaction of shrimp DNA and T-2 toxin in vitro is verified the above experimental results.In the end, using T-2 toxin feed the shrimp for 20 days, the effects of T-2 toxin on the DNA viscosity were observed. Results:the response surface analysis of optimum reaction conditions were the most obvious impact on the DNA viscosity, the conditions of T-2 toxin is 2.7ng/mL; the conditions of DNA is 50μg/mL; the reaction time is 43min. The reactions of T-2 toxin on calf thymus DNA and shrimp DNA in vitro induced consistent changes of DNA viscosity. The DNA viscosity of the T-2 toxin in the dose group was not completely consistent with the experimental results in vitro.
