不同条件对勐库大叶种多酚氧化酶合成茶黄素的影响

摘要: 酶促合成茶黄素一直是国内外研究的热点,但缺乏对云南大叶种多酚氧化酶催化合成茶黄素的研究。本研究分析了不同因素对勐库大叶种多酚氧化酶催化合成茶黄素的影响,结果表明在pH4.0、37 °C、茶多酚浓度为4.5 mg/mL的条件下,勐库大叶种多酚氧化酶催化1.0 h,最有利于茶黄素的合成,其总产量可达461.12±15.01 μg/mL,转化率为15.31±0.40%。勐库多酚氧化酶催化合成酯型茶黄素为主,对酯型儿茶素活性更强,在催化过程中还伴随着EGC和GA的生成,有利于茶黄素的合成积累,这可能与滇红优异品质形成有关。

Abstract: Enzymatic synthesis of theaflavins has attracted more and more attention. However, there were no reports on the enzymatic synthesis of theaflavins by polyphenol oxidase (PPO) from C. sinensis var. Mengku. So the effects of different factors on the synthesis of theaflavins by crude PPO from C. sinensis var. Mengku were analyzed. The results showed that the theaflavins yield of C. sinensis var. Mengku crude PPO reached 461.12±15.01 μg/mL under the optimized catalytic parameters of pH4.0, 37 °C of reaction temperature, 4.5 mg/mL of substrate concentration, and 1 h of reaction time, and the rate of theaflavins conversion was 15.31±0.40%. Additionally, ester theaflavins are the major product of crude PPO from C. sinensis var. Mengku. And EGC and GA were generated during PPO oxidation, which benefits the synthesis and accumulation of theaflavins. These results may be the reasons resulting in high quality of Dianhong.
