蔷薇红景天低聚体原花青素对动脉粥样硬化大鼠肝脏的保护作用

The Protective Effect of Oligomeric Procyanidins from Rhodiola Rosea L.on the Liver of Atherosclerosis Rats

摘要: 目的:研究蔷薇红景天低聚原花青素(OPCRR)对动脉粥样硬化(AS)大鼠肝脏的保护作用。方法:采用维生素D3结合高脂乳剂灌胃的方法建立大鼠动脉粥样硬化模型。随机分为阴性对照组、模型对照组、OPCRR 60、120、240 mg/kg?bw剂量组和姜黄素(50 mg/kg?bw)对照组。连续灌胃8周后,通过分析各组动物的肝脏的组织形态、脂质、氧化应激和炎症细胞因子等指标的变化,研究OPCRR对大鼠动脉粥样硬化的干预效果。结果:与AS模型组相比,OPCRR可以显著降低大鼠血清中TC、TG、LDL-c水平,显著提高HDL-c水平,并降低大鼠AI数值(P<0.05,P<0.01);OPCRR可显著减少肝脏中的炎性灶和细胞变性,减少肝细胞的受损程度;肝组织TC、TG、LDL-c和VLDL-c水平显著下降,HDL-c水平显著升高(P<0.05,P<0.01);肝脏中MDA的含量显著减少,SOD和GSH-Px的活力显著提高(P<0.05,P<0.01);同时肝脏中的IL-1β、IL-6、IL-10、TNF-α、MCP-1和 ICAM-1水平显著下降(P<0.05,P<0.01)。结论:蔷薇红景天低聚原花青素可以通过改善肝脏中脂质代谢、氧化应激和炎症反应而起到对AS大鼠的肝脏的保护作用,且整体效果优于姜黄素。

Abstract: Objective: This experiment mainly aimed to study the effect and possible mechanism of ability of oligomeric procyanidins from Rhodiola rosea L.on the liver of atherosclerosis rats. Methods:Atherosclerotic model was established by lavaging with a loading does of vitamin D3 and high-fat emulsion. The animals were divided into following group, control, As, curcumin (50mg/kg), OPCRR 60 mg/kg?bw,120 mg/kg?bw and 240 mg/kg?bw groups. After 8 weeks, we determined the liver tissue pathology change, lipid, oxidative stress and inflammatory cytokines. Results: compared with the AS model group, OPCRR significantly reduced the levels of TC, TG, and LDL-c in the serum, significantly raising the level of HDL-c in the serum, at the same time, OPCRR can also significantly reduced the number of AI of rats,OPCRR can significantly improve the morphology of liver cells, protect the structure of hepatic lobule, significantly reduce inflammatory foci and cell degeneration in the liver and reduce the damage rate of the liver cells; The TC, TG, LDL-c and VLDL-c level in liver dropped significantly, HDL-c were significantly increased (P < 0.05, P < 0.01); content of MDA in the liver significantly reduced, the activities of SOD and GSH-Px increased significantly (P < 0.05, P < 0.01); At the same time the inflammatory cytokines levels in the livers (IL-1 beta, IL-6, IL-10, the TNF alpha, MCP-1 and ICAM-1) decreased significantly (P < 0.05, P < 0.01). Conclusion:OPCRR can modify liver lipid metabolism, oxidative stress and inflammation and have the protective effect on the AS rat liver, and the effect is better than those of curcumin.
