枸杞对线虫寿命和产卵的影响及其抗氧化作用的研究

Effects of Lycium Barbarum on longevity and spawning of Nematode and its mechanism of antioxidant capacity

摘要: 摘要:目的:研究枸杞对线虫寿命和产卵的影响,探讨其抗氧化的相关机制。方法:利用FRAP法体外检测比较了宁夏中宁、青海、甘肃三个产地的枸杞抗氧化能力的差异。采用ROS-敏感染料DCFH-DA检测枸杞对线虫体内ROS的影响,同时采用实时定量荧光PCR的方法检测枸杞对线虫体内相关抗氧化酶水平的影响。同步化后L4时期的线虫分别放置于对照或枸杞接种的NGM平板上培养,检测枸杞对线虫产卵和寿命的影响。采用Pmyo3 :: HyPer和roGFP2 :: Orp1两种转基因线虫为模型检测枸杞提取物对线虫胞质中H2O2的清除能力,采用O2?·荧光染料MitoSox Red检测枸杞提取物对线虫细胞线粒体中超氧阴离子(O2?·)的清除能力。结果:三种产地的枸杞均具有很强的体外抗氧化能力,它们之间没有显著差异。枸杞可以降低线虫体内ROS水平以及提高抗氧化酶的水平。枸杞对线虫寿命和产卵能力并没有显著的影响。枸杞可以清除线粒体超氧阴离子但不影响胞质H2O2水平。结论:枸杞在体外与体内均有一定的抗氧化能力,主要通过清除线粒体超氧阴离子在体内发挥抗氧化作用。枸杞对线虫的寿命和产卵数没有显著影响。

Abstract: Abstract: Objective: It was reported that Lycium barbarum had antioxidant and anti-aging ability, our experiments aimed to study the effects of Lycium barbarum on the life span and spawning of nematodes, and to explore the mechanism of its antioxidant capacity. Methods: FRAP method was used to compare the antioxidant capacity of Lycium barbarum from three different areas of Ningxia, Qinghai and Gansu provinces. To detect the effect of Lycium barbarum in vivo, the nematodes of L4 stage were placed on the NGM plates incubated with or without Lycium barbarum, and the lifespan and the number of egg laid was analyzed. The ROS levels of nematodes were detected using the ROS-sensitive dye DCFH-DA, and the expression of antioxidant enzymes was evaluated by Real-time Quantitative PCR. The effects of Lycium barbarum on the scavenging of H2O2 in cytoplasm were detected using two transgenic nematodes: Pmyo3 :: HyPer and roGFP2 :: Orp1. MitoSox Red was used to measure the superoxide anion in mitochondria. Results: There was no significant difference of the antioxidant abilities of the three kinds of Lycium barbarum. And in vivo, Lycium barbarum could reduce the level of ROS and increase the levels of antioxidant enzymes. However, Lycium barbarum has no significant effect on nematode longevity and spawning ability. To further investigate which specific type of ROS was influenced by Lycium barbarum, we found that the level of mitochondrial O2?· but not cytoplasmic H2O2 was reduced. Conclusion: Lycium barbarum exerts antioxidant capacity in vitro and in vivo, and its effect in vivo is achieved by reducing the level of mitochondrial O2?·. In addition, Lycium barbarum does not dramatically affect the lifespan and the total number of eggs laid in nematodes.
