采后BTH处理通过激活苯丙烷代谢和抗氧化酶促进梨果实的愈伤

Postharvest BTH Treatment Promotes Wound Healing of Pear Fruit by Activating Phenylpropane Metabolism and Antioxidant Enzymes

摘要: 摘要:【目的】研究苯丙噻重氮(benzothiadiazole,BTH)处理对梨果实采后愈伤的促进作用及部分机理。【方法】以“玉露香”梨果实为试材,人工模拟损伤后用100ppm BTH处理,在常温(20-25℃,RH80-85%)黑暗条件下进行愈伤,通过分析愈伤期间苯丙氨酸解氨酶、超氧化物歧化酶、过氧化物酶和多酚氧化酶的活性,以及总酚、类黄酮和木质素的含量来评价愈伤效果。【结果】BTH处理对果实愈伤期间的失重率没有显著影响,但显著降低了果实的发病率,处理后4d,果实的发病率仅为对照果实的40%,由此表明,BTH处理促进了梨果实的采后愈伤。BTH处理还明显提高了果实苯丙氨酸解氨酶、多酚氧化酶、过氧化物酶和超氧化物歧化酶的活性,处理后12h,处理果实的苯丙氨酸解氨酶和超氧化物歧化酶活性分别高出对照1.29倍和50%;处理后1d,处理果实的多酚氧化酶和过氧化物酶活性分别高于对照果实6.4倍和2.56倍。此外,BTH处理还有效促进了果实总酚、类黄酮和木质素的积累,处理后1d,处理果实的总酚和类黄酮含量分别高出对照19%和63%,处理后12h,处理果实的木质素含量高出对照1.4倍。相关性分析结果表明,处理果实的发病率和PAL活性、总酚含量和类黄酮含量之间呈显著负相关,和POD及PPO活性之间呈极显著负相关。【结论】BTH处理可以通过诱导果实的苯丙烷代谢,提高抗氧化酶的活性来促进梨果实的采后愈伤。

Abstract: Abstract: [Objective] To study the promotion effect of benzothiadiazole (BTH) treatment on wound healing of harvested pear fruit. [Method] Pear cultivar ‘Yuluxiang’ was employed as material. The fruit was artificially damaged, tread with BTH at 100ppm, and wound healing under dark condition at ambient temperature (20-25oC, RH80-85%). The healing effects were evaluated by determining the activity of phenylalanine ammonia lyase (PAL), superoxide dismutase (SOD), peroxidase (POD) and polyphenol oxidase (PPO), and the content of total phenolics, flavonoids and the lignin. [Results] BTH treatment had no significant effect on the weight loss, however, reduced significantly the incidence of inoculated fruit during healing, the incidence of treated fruit was only 40% of the control fruit after 4 days of treatment. The result indicated BTH promotes wound healing of harvested pear fruit. The treatment also significantly increased the activity of PAL, SOD, POD and PPO of fruit, the activity of PAL and SOD was 1.29 times and 50% higher than the control after 12h treatment, and. the activity of PPO and POD was 6.4 times and 2.56 times higher than the control after 1 days of treatment. In addition, the chemical effectively promoted the accumulation of total phenols, flavonoids and the lignin, the content of total phenolics and flavonoids was 19% and 63% higher than the control after 1day of treatment, the contend of lignin was 1.4 times higher than the control after 12h of treatment. The correlation analysis showed that there was a significant negative relationship between incidence and PAL activity, flavonoid contents in treated fruit .[Conclusion] BTH treatment promotes wound healing of harvested pear fruit by activating phenylpropane metabolism and antioxidant enzymes.
