绞股蓝中6种多酚化合物的超高效液相色谱检测及其醇提液的抗氧化活性研究

Studies of ultra performance liquid chromatography method to determination of six phenolic compounds in Gynostemma pentaphyllum Makino and the antioxidant activity of methanol extracts

摘要: 超高效液相色谱法(UPLC)分析五叶、七叶、九叶、和人工种植的4种绞股蓝的叶及枝中6种多酚化合物的含量,并测定样品醇提液的总多酚含量及抗氧化活性。结果表明:色谱柱为poroshell 120 PFP(4.6 mm×100 mm,2.7 μm),流动相为1%甲酸水(A)和乙腈(B),检测波长为254 nm和 350 nm的条件下,绞股蓝样品中6种多酚单体可在35 min内得到较好分离,且重复性好(RSD≤4.6%)、精密度高(RSD≤2.68%)、稳定性好(RSD≤2.45%)、加标回收结果准确可靠(平均回收率84%~99%,RSD≤4.02%)。绞股蓝中6种多酚化合物、总多酚及抗氧化活性在不同品种间差异较大。其中,芦丁在绞股蓝中含量较高,对羟基苯甲酸含量较低。绞股蓝醇提液中总多酚及抗氧化活性在不同品种间差异较大,在七叶绞股蓝的叶中最高,在七叶的枝中最低。

Abstract: A new ultra performance liquid chromatography (UPLC) was used to analyze the content of 6 phenolic compounds in the leaves and branches of five kinds of Gynostemma pentaphyllum Makino, five-leaf, seven-leaf, nine-leaf, and artificially planted. And the total polyphenols content and the antioxidant activity of the methanol extract of the samples were determined. The column was poroshell 120 PFP (4.6 mm×100 mm,2.7 μm) and the mobile phase was 0.01% formate water (A) and acetonitrile (B), and the detection wavelength was 254 nm and 350 nm. The results showed that 6 phenolic compounds in Gynostemma pentaphyllum Makino could be separated well within 35 min, with good repeatability (RSD≤4.6%), high precision (RSD≤2.68%), good stability (RSD≤2.45%) and reliable recovery (average recovery rate 84%~99%, RSD≤4.02%). The six phenolic compounds, total polyphenols and antioxidant activities in Gynostemma pentaphyllum Makino varied greatly among different varieties. Among them, the rutin content in Gynostemma pentaphyllum Makino was higher than other phenolic compounds and the p-hydroxybenzoic acid content was lower others. The total polyphenols content and antioxidant activity in leaves of seven-leaf Gynostemma pentaphyllum Makino were the highest among the 8 samples, and the lowest was in the branch of seven-leaf.
