巨大芽孢杆菌L2提取物对魔芋软腐病菌的抑菌机制研究

Antagonistic bacteria Mechanism of extraction from Bacillus megaterium L2 against Erwinia carotovora

摘要: 本试验旨在探究巨大芽孢杆菌(Bacillus megaterium L2)提取物对贮藏期魔芋软腐病原菌——胡萝卜软腐欧文氏菌(Erwinia carotovora EC-1)的抑菌机制。以巨大芽孢杆菌菌粉为原料,用常压硅胶柱层析分离得到提取物LE4-1~LE4-7,经抑菌实验得到对胡萝卜软腐欧文氏菌EC-1有良好抑菌作用的提取物,再使用气相色谱-质谱(GC-MS)对其进行成分分析;并通过测定其半数抑制浓度IC50值(half maximal inhibitory concentration)、菌液电导率、核酸蛋白泄露、可溶性总糖含量、菌体总蛋白、菌体磷代谢及胞内活性氧(ROS)含量对其抑菌机制进行研究。结果表明:巨大芽孢杆菌L2提取物LE4-3对EC-1有良好抑菌作用,经气相色谱-质谱(GC-MS)检测其主要成分为苯乙酸甲酯(32.19%)、亚油酸乙酯(15.819%)、苯乙酸乙酯(13.793%)等;提取物LE4-3对EC-1的IC50值为1.06 mg/mL,并初步推测其作用机制是通过影响细胞膜的完整性、损伤细胞结构、抑制细胞蛋白质合成及能量代谢等方面对EC-1发挥抑菌作用,因此巨大芽孢杆菌提取物LE4-3在魔芋贮藏期软腐病的抗病贮藏中具有良好的应用前景。

Abstract: The aim of this experiment was to investigate the inhibitory mechanism of Bacillus megaterium L2 extract against Erwinia carotovora EC-1 of soft rot bacteria in Amorphophallus konjac during storage. The extract LE4-1~LE4-7 was isolated by silica gel column chromatography under atmospheric pressure from Bacillus megaterium powder , and the extract with good bacteriostatic effect on Erwinia carotovora EC-1 was obtained by bacteriostatic experiment, and it’s Composition was analyzed by gas chromatography-mass spectrometry (GC-MS).The bacteria mechanism was studied by measuring the IC50 value (half maximal inhibitory concentration), nucleic acid protein leakage, soluble total sugar content, cell total protein, phosphorus metabolism and reactive oxygen species (ROS).The results showed that the extract LE4-3 had a good bacteriostatic effect on EC-1 from Bacillus megaterium L2, and its major compounds included methyl phenylacetate (32.19%),ethyl linoleate (15.819%),ethyl phenylacetate (13.793%),ethyl stearate (9.105%).The IC50 value of the extract LE4-3 on EC-1 was 1.06 mg/mL, and the antimicrobial mechanism may be achieved by damaging bacterial cell structure, affecting the integrity of cell membrane,reducing protein content and energy metabolism until the EC-1 are broken into pieces. Therefore, the extract LE4-3 has a good application prospect in the Amorphophallus konjac storage of Erwinia carotovora of soft rot bacteria from Bacillus megaterium L2.
