食(药)用蕈菌中高抑菌活性菌株筛选及培养基优化

Screening of Strains with High Antibacterial Activity from Edible and Medicinal Fungi and Optimization of Culture Medium

摘要: 对4个不同属24株食(药)用蕈菌对11株指示菌抑菌活性进行研究,通过初筛模型,从中筛选出3株功能菌,6株指示菌。对初筛菌株进一步复筛,确定功能菌株H2与枯草芽孢杆菌、沙门氏菌、蜡状芽孢杆菌为培养基优化的出发菌株和指示菌。通过响应面分析优化培养基配方,结果表明,当麦芽糖40.9g/L、蛋白胨11.2g/L、KH2PO4 2.1g/L、MgSO4 ·7H2O 1.1g/L时,H2对枯草芽孢杆菌、沙门氏菌、蜡状芽孢杆菌的抑菌活性均较理想,其抑菌圈半径分别达到3.513、4.610、8.800mm。

Abstract: The antibacterial activities of 24 edible or medicinal fungus strains belonging to 4 genera against 11 indicator bacteria were studied. Three edible fungi with obvious antibacterial activity and 6 indicator bacterial strains were preliminarily screened. Secondary screening was carried out to pick out strain H2 and three indicator bacteria including Bacillus subtilis, Salmonella typhi, and Bacillus cereus for further research into the optimization of culture medium for strain H2 using response surface methodology based on inhibitory zone radii against the indicator bacteria. The results showed that the optimal culture medium for strain H2 was composed of 4.09% maltose, 1.12% peotone, 0.21% KH2PO4, and 0.11% MgSO4 ·7H2O. The inhibitory zone radii of the strain against B. subtilis, S. typhi and B. cereus were 3.513, 4.610 mm and 8.800 mm, respectively after culture in the medium.
