扫描电镜法分析荞麦蜂花粉破壁及其黄酮模拟胃肠液吸收

Scanning electron microscope analysis of the wall-breaking of buckwheat bee pollen and its flavonoids absorption in simulated gastrointestinal fluid

摘要: 摘要:本文以扫描电镜法研究了荞麦蜂花粉低温聚能超声破壁过程,发现花粉破壁是在外加聚能超声作用下,沿三个萌发沟裂开、接着破裂,最后破碎的过程,据此提出了电镜视野中更加精准的破壁率计算方法。在电镜观测指导下,获得了最佳的破壁条件为:初始温度0℃,超声功率400 W,脉冲通断比为1:2,超声时间30 min,料液比1:30,在此条件下,破壁率达98.33%。进一步研究发现,破壁率与蜂花粉细胞内黄酮在模拟肠胃液中的总吸收率呈线性增加关系(相关系数R2=0.9997)。

Abstract: Abstract: Low temperature polymeric ultrasound was used in the breaking wall of buckwheat bee pollen and the wall breaking process was analysis using scanning electron microscope(SEM). The bee pollen cells were splited, ruptured along the germ ditch, and broken to into pieces successively in SEM field. A more accurate calculation method for wall breaking rate in SEM field was proposed. Based on SEM observation, a ratio of wall-broken 98.33% was obtained under the best conditions for breaking the cell wall: initial temperature 0 oC, ultrasound power 400 W, the pulse duty factor was 1:2, ultrasound time 30 min, the ratio of material to liquid 1:30. Further study shows that the wall-cracking rate bears a linear relationship to the total absorptance rate of bee pollen flavone in simulated gastric and intestinal fluid (the correlation coefficient R2=0.9997) .
