两种饲养方式下苏尼特羊肉中的鲜味物质及相关调控基因表达量的研究

Effects of Two Feeding Patterns on umami substances and related genes of Sunit Sheep Meat

摘要: 以舍饲、放牧两种饲养方式下的12 月龄苏尼特羊不同部位的肌肉为实验材料,取股二头肌、背最长肌、臂三头肌3个部位,分别测定味觉指标、鲜味物质以及相关调控基因并进行比较分析。结果表明:放牧的苏尼特羊肉鲜味和咸味高于舍饲,苦味和涩味低于舍饲;在鲜味物质中,放牧条件下苏尼特羊肉中的肌苷酸(inosinic acid, IMP)和肌苷(inosine, INO)含量高于舍饲;放牧方式对次黄嘌呤(Hypoxanthine, HPY)含量没有显著影响,但均在背最长肌中含量最高(P<0.05);放牧条件下背最长肌的腺苷一磷酸脱氨酶(AMPD1)基因表达量显著高于舍饲(P<0.05),股二头肌的腺苷酸琥珀酸裂解酶(ADSL)和次黄嘌呤核苷酸环水解酶(ATIC)基因表达量显著高于舍饲(P<0.05);鲜味物质调控基因的相关性中,放牧羊肉AMPD1-ADSL之间的正相关达到极显著性水平(P<0.01);通过分析鲜味物质与调控基因相关性发现,放牧条件下IMP-AMPD1基因之间呈极显著正相关(P<0.01),IMP-ATIC基因之间呈显著正相关(P<0.05),舍饲条件下IMP-AMPD1基因之间呈显著正相关(P<0.05),相关性结果验证了当鲜味物质调控基因的表达量高时,肉中的鲜味物质含量也随之提高。

Abstract: The objective of this study was to investigate the difference in taste characteristics, umami substances and umami related genes of Longissimus dorsi, Biceps femoris and Triceps brachii muscles from Sunit sheep subjected to two feeding patterns (grazed and concentrated). Sunit sheep were slaughtered at 12 months of age for the collection of muscle samples. The results showed that umami and saltiness in grazed were higher than concentrated, while bitterness and astringency were lower than concentrated. Among the umami substances, the IMP and INO content in grazed was higher than concentrated. Feeding patterns had no significant effect on HPY, however HPY were all highest in longissimus muscle (P<0.05). The expression of AMPD1 from grazed sheep were significantly higher than the concentrated in Longissimus dorsi (P>0.05), the expression of AMPD1 from grazed sheep were significantly higher than the concentrated in Biceps femoris (P<0.05). In grazed conditions, the expression of AMPD1 had greatly significant positive correlated with ADSL (P<0.01). Correlation analysis showed that IMP was positively correlated with AMPD1 (P<0.01), ATIC (P<0.05) in grazed sheep; IMP was positively correlated with AMPD1 (P<0.05) in concentrated sheep. The correlation results confirmed that when the expression level of IMP-regulated genes is high, The IMP content also increases.
