甜菜红素对小鼠体内抗氧化及免疫调节的作用

Antioxidant Activity and Immunomodulatory Effect of Betalain in Mice

摘要: 目的:研究甜菜红素对KM小鼠体内抗氧化及免疫调节的作用。方法:利用制备的甜菜红素作为供试品;动物实验分为空白组及甜菜红素高、中、低3 个剂量组,对雄性KM小鼠连续灌胃15 d,测定各组动物血清超氧化物歧化酶(superoxide dismutase,SOD)活力、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)活力及丙二醛(malondialdehyde,MDA)浓度;测定各组动物免疫器官脏器指数、脾淋巴细胞增殖能力、血清溶血素水平、自然杀伤(natural killer,NK)细胞活性及腹腔巨噬细胞吞噬功能的变化情况。结果:与空白组相比,高、中剂量的甜菜红素可以显著提高小鼠血清中SOD、GSH-Px活力(P<0.05),显著降低MDA浓度(P<0.05);高、中、低剂量组小鼠免疫器官胸腺指数显著高于空白组(P<0.05),高、中剂量组小鼠的脾脏指数、吞噬指数显著高于空白组(P<0.05),脾淋巴细胞增殖能力、血清溶血素水平和NK细胞活性与空白组比较有显著差异(P<0.05)。结论:甜菜红素能显著提高小鼠体内抗氧化活性及特异性和非特异性免疫能力。

Abstract: Objective: To explore the regulatory effect of betalain on antioxidant status and immune function in mice. Methods: Betalain was extracted and separated from red beet (Beta vulgaris var. rubra). Three groups of male KM mice were used: blank group and high, medium and low-dose betalain groups. After 15 days of oral administration, the activity of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px), and the concentration of malondialdehyde (MDA) in serum were determined in each group. The changes in immune organ index, splenic lymphocyte proliferation, serum hemolysin, natural killer (NK) cell activity and the phagocytic function of peritoneal macrophages were measured. The results showed that the activities of SOD and GSH-Px in the serum of mice were significantly increased in the high and middle-dose groups and MDA concentration was significantly decreased compared with the blank group (P < 0.05). The thymus index of mice in each dose group was higher than that in the blank group (P < 0.05), and the spleen index and corrected clearance index of mice in the high and medium-dose groups were both significantly higher than those in the blank group (P < 0.05). Also, the splenic lymphocyte proliferation ability, serum hemolysin and NK cell activity in the two dose groups were significantly different from those in the blank group (P < 0.05). It was concluded that betalain could significantly improve antioxidant status and specific and nonspecific immunity of mice.
