婴儿源动物双歧杆菌消化道胁迫抗性及体外对免疫细胞活性影响的比较研究

Comparison of resistance of Bifidobacterium isolated from infants to simulated gastrointestinal stress and its effect on immune cell activity in vitro

摘要: 以4株婴儿源动物双歧杆菌乳酸亚种为研究对象,分析了菌株对酸、胆盐、模拟消化道环境的耐受能力及对致病菌的黏附抑制作用,并通过对Caco-2细胞的黏附评价其黏附性。此外,还测定了菌株合成胞外多糖的能力,并探讨了其与耐受性及黏附性的关联。经上述实验获得的黏附性及抗性双优的菌株H15-2,再经脾淋巴细胞增殖试验、巨噬细胞能量代谢水平及巨噬细胞吞噬中性红能力三个方面评价其免疫调节能力。结果表明:动物双歧杆菌H15-2具有良好的耐受性及黏附能力,并对免疫细胞活性具有较好的调节作用,有应用开发潜力。

Abstract: Four strains of Bifidobacterium animalis subsp. lactis isolated from infants were analyzed including the tolerance to acids, bile salts, simulated digestive tract environment and the inhibitory effects to pathogen adhesion. The adhesive ability of the strains was evaluated by adhesion to Caco-2 cells in vitro. In addition, the abilities of exopolysaccharides synthesis were also determined and the relationship between tolerance and adhesion was discussed. The strain H15-2 with both of good tolerance and adhesion was evaluated for its immunomodulatory ability which included splenocyte proliferation, energy metabolism level of macrophage, and macrophage phagocytosis of neutral red. The results showed that Bifidobacterium animalis H15-2 had strong tolerance to simulated gastrointestinal environment, high adhesion ability to Caco-2 cells, and good regulating effect on immune cell activity. It has potential for application development.
