牛磺酸对抑郁症模型小鼠的干预作用

摘要: 摘要:目的:研究牛磺酸对抑郁的干预作用。方法:采用行为绝望和利血平诱导的急性抑郁症两种小鼠模型。随机分成6组:阴性对照组、模型对照组、氟西汀对照组、三个牛磺酸剂量组(0.5、1、2 g/ kg?bw),每组10只。通过悬尾实验、强迫游泳实验、旷场实验建立的行为绝望小鼠模型进行研究,研究牛磺酸对小鼠到达行为绝望时间的影响;通过分析眼睑下垂、肛温、出圈率等行为学指标,血清中促肾上腺皮质激素(Adreno-Cortico-Tropic-Hormone ,ACTH)、脑源性神经营养因子(Brain Derived Neurotrophic Factor ,BDNF)、皮质醇(Cortisol ,COR),脑组织中5-羟色胺(5-hydroxytryptamine ,5-HT)、去甲肾上腺素(Norepinephrine ,NE)、多巴胺(Dopamine ,DA)、单胺氧化酶(Monoamine Oxidase, MAO)、促肾上腺皮质激素释放因子(Corticotropinreleasing Facto,CRF)等生化指标,研究牛磺酸对急性抑郁的干预作用。结果:牛磺酸显著性缩短了行为绝望模型小鼠悬尾不动时间(P<0.01)和强迫小鼠游泳不动时间(P<0.01),在旷场实验中,显著性升高水平、垂直运动次数(P<0.01),眼睑下垂度无显著性差异(P>0.05),显著升高抑郁小鼠脑组织5-HT、DA、NE水平(P<0.05),降低MAO活性(P<0.01),并升高BDNF含量(P>0.05),同时抑制CRF、ACTH、COR的过度分泌(P<0.01),并改善相关生化指标。结论:牛磺酸可能是通过改善模型动物的激素分泌干预利血平诱导的急性抑郁症。

Abstract: Abstract: Objective: To study the intervention effect of taurine on depression. Methods: Two mouse models of acute depression induced by behavioral despair and reserpine. They were randomly divided into 6 groups: negative control group, model control group, fluoxetine control group, and three taurine dosage groups (0.5, 1, 2 g/kg?bw), with 10 rats in each group. Through the behavioral despair mouse model established by tail suspension experiment, forced swimming experiment and open field experiment, the effect of taurine on the despair time of mice was studied. The behaviors such as drooping, rectal temperature and outlet rate were analyzed. Indicators, serum ATCH, BDNF, COR, brain tissue 5-HT, NE, DA, MAO, CRF and other biochemical indicators, to study the intervention of taurine on acute depression. RESULTS: Taurine significantly shortened the time of suspension of the behavioral despair model mice (P<0.01) and forced swimming time (P<0.01). In the open field experiment, the level of significant increase, The number of vertical movements (P<0.01), there was no significant difference in eyelid sag (P>0.05), significantly increased the levels of 5-HT, DA, NE in brain tissue of depressed mice (P<0.05), and decreased MAO activity (P< 0.01), and increased BDNF content (P>0.05), while inhibiting excessive secretion of CRF, ACTH, COR (P<0.01), and improved related biochemical indicators. Conclusion: Taurine may be an intervention in reserpine-induced acute depression by improving hormone secretion in model animals
