虾蛄不同可食组织中镉和砷的形态及分布特征

Species analysis and the distribution characteristic of cadmium and arsenic in the different edible tissues of Oratosquilla oratoria

摘要: 为了揭示虾蛄可食组织中镉和砷的形态及分布特征,分别运用电感耦合等离子体质谱法和高效液相色谱-电感耦合等离子体质谱联用技术分析虾蛄不同可食组织(肌肉和性腺)中镉和砷的总量及其形态。结果显示,虾蛄对镉具有极强的富集能力,实验采集的样品可食组织中总镉超标率达100%且总镉的含量与雌雄性别无相关性,其中性腺组织中总镉的含量约为肌肉组织的10 倍,形态分析显示大部分样品中毒性最高的无机离子态镉占总镉含量低于20%,且各组织中的无机离子态镉与总镉呈显著正相关关系。虾蛄可食组织中总砷含量较高且与雌雄性别无相关性,性腺组织中的总砷含量普遍高于肌肉组织。实验采集的所有虾蛄样品可食组织中均检出三价无机砷和五价无机砷,部分样品和组织中检出一甲基砷和二甲基砷,其中肌肉组织中无机砷含量最高为0.46 mg/kg,未超出国家标准限量值,无机砷占总砷量为1.71%~6.14%,可食组织中无机砷的含量与总砷之间无显著相关关系。砷甜菜碱是虾蛄可食组织中的主要砷形态。因此针对虾蛄高富集镉和砷的特性,亟需在形态分析的基础上深入开展镉和砷的生物利用率研究,提出科学、合理的标准限量值。

Abstract: Present study was aimed to investigate the distribution characteristic and the species of cadmium and arsenic in the edible tissues of O. oratoria. Inductively coupled plasma mass spectrometry (ICP-MS) was used for measurement of the total content of arsenic and cadmium in the edible muscle and gonad, and the high performance liquid chromatography-inductively coupled plasma mass spectrometry (HPLC-ICP-MS) for analyzing the free inorganic Cd2+ and different species of arsenic. Results showed that O. oratoria could accumulate high concentrations of cadmium in the edible tissues, and the total cadmium concentration in all samples collected in present study had exceeded the maximum permitted concentration (MPC) of 0.50 mg kg-1 Cd wet weight (w. w.) in our country. In addition, the total cadmium concentration had no significant relationship with female or male. And the total cadmium in gonad was almost 10 times of the level in muscle. Species analysis showed that the percent of the most toxic free inorganic Cd2+ to the total cadmium was below 20% for most of the samples, and there was a significant positive relationship between the inorganic Cd2+ and the total cadmium. The total arsenic concentration in the edible tissues of O. oratoria were high, which was proved no significant relationship with different sex. However, the concentration of total arsenic in the gonad was higher than that in the muscle. The inorganic arsenic was detected in all samples, and MMA and DMA were also found in some samples. The highest concentration of inorganic arsenic was 0.46 mg/kg in muscle which were below the maximum standard limitation of our country, and the percentage of inorganic arsenic to total arsenic in muscle ranged from 1.71% to 6.14%. In addition, there was no significant relationship between inorganic arsenic and total arsenic for all the edible tissues. The organic arsenic AsB was the main species for the muscle and gonad of O. oratoria. Because of the high accumulation capacity for total cadmium and arsenic, a more accurate maximum permitted concentration (MPC) of Cd for the edible tissues in O. oratoria is urgent to be established and the metal bioaccessibility and bioavailability must be considered to improve the risk assessment analysis.
