CaCO3对凝结芽孢杆菌CGMCC 9951芽孢形成的影响

Effect of CaCO3 on sporulation of Bacillus coagulans CGMCC9951

摘要: 凝结芽孢杆菌是一种兼具乳酸菌和芽孢杆菌特点的重要益生菌之一,近年来受到越来越广泛关注,但其芽孢形成途径及调控机制的研究却相对匮乏。本文主要研究了碳酸钙添加对凝结芽孢杆菌CGMCC 9951芽孢形成的影响及可能机制。结果表明,在16 h添加6%碳酸钙后,不仅确保了芽孢形成时充足的钙要求,还可促使发酵液pH由4左右迅速提升至6.5以上,快速启动芽孢的形成。在培养68±2 h时,芽孢形成量可达4.53×107 CFU/mL,比对照组芽孢化启动提前约100 h,且芽孢耐热性、耐酸、耐胆盐及其萌发能力均未受显著影响。进一步研究发现,碳酸钙的添加有效加速了凝结芽孢杆菌CGMCC 9951芽孢形成进程,在添加碳酸钙4 h后芽孢化启动相关基因kinC、kinE、spo0A表达量显著提高,分别为对照组的61.07倍、23.49倍、17.32倍。该研究初步揭示凝结芽孢杆菌芽孢形成具有一定的环境pH依赖性,中性偏碱的环境有利于芽孢化启动相关基因的转录,进而加速芽孢的形成过程。

Abstract: Bacillus coagulans is one of the most important probiotics with the characteristics of both lactic acid bacteria and Bacillus. In recent years, it has attracted more and more attention. However, the study on the sporulation pathway and regulation mechanism of Bacillus coagulans is relatively scarce. The effect of calcium carbonate addition on sporulation of Bacillus coagulans CGMCC 9951 and its possible mechanism were mainly studied in this paper. The results showed that the addition of 6% CaCO3 at 16 h not only ensured sufficient calcium requirement for sporulation, but also promoted the pH of fermentation broth from about 4 to over 6.5, and quickly initiated sporulation. At 68±2 h, the sporulation amount reached 4.53×107 CFU/mL, which sporulation initiation time was about 100 hours earlier than the control group, and the heat resistance, acid resistance, bile salt tolerance and germination ability of the spores were not significantly affected. Further studies found that the addition of CaCO3 effectively promoted the sporulation process of Bacillus coagulans CGMCC 9951. The expression of kinC, kinE and spo0A which related to sporulation initiation was significantly increased after adding CaCO3 for 4 h, which was 61.07 times, 23.49 times, 17.32 times of the control group, respectively. This study preliminarily revealed that the sporulation of Bacillus coagulans had a certain environmental pH dependence, and the neutral alkaline environment was conducive to the sporulation initiation of gene transcription, thus accelerating the sporulation process.
